目的:研究灯盏细辛注射液(Erigeron breviscapusinjection,EBI)对离体培养的大鼠脑微血管内皮细胞(BMEC)炎症相关细胞因子白细胞介素-1β(IL-1β)及细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)蛋白表达的影响,初步探讨EBI抗脑缺血炎症损伤的机制。方法:采用肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)建立离体培养的大鼠BMEC炎症损伤模型,分别用0.1 mg.L-1和1 mg.L-1EBI处理内皮细胞,使用ELISA试剂盒分析IL-1β蛋白表达的变化,流式细胞仪测定ICAM-1和VCAM-1荧光强度的变化。结果:EBI可以抑制TNF-α诱导的内皮细胞IL-1β高表达,降低TNF-α诱导的内皮细胞ICAM-1和VCAM-1高表达。结论:EBI可以降低由TNF-α诱导的内皮细胞过度表达IL-1β,ICAM-1和VCAM-1,从而对脑缺血炎症损伤具有一定保护作用。
Objective: To investigate the effects of Erigeron breviscapus injection (EBI) on inflammatory cytokines IL-1β and ICAM-1 in rat brain microvascular endothelial cells (BMECs) (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) protein expression, and to explore the mechanism of anti-ischemic inflammation in EBI. Methods: The inflammatory injury model of rat BMECs cultured in vitro was established by using tumor necrosis factor-α (TNF-α). Endothelial cells were treated with 0.1 mg · L-1 and 1 mg · L-1 EBI, The changes of IL-1βprotein expression were analyzed by ELISA kit, and the changes of fluorescence intensity of ICAM-1 and VCAM-1 were measured by flow cytometry. Results: EBI could inhibit TNF-α-induced high expression of IL-1β in endothelial cells and decrease the expression of ICAM-1 and VCAM-1 in endothelial cells induced by TNF-α. Conclusions: EBI can reduce the over-expression of IL-1β, ICAM-1 and VCAM-1 induced by TNF-α in endothelial cells, which may have a protective effect on inflammatory injury of cerebral ischemia.
