目的:利用神经元特异性烯醇化酶(NSE)免疫组化染色,光镜下观察慢性砷中毒对成年大鼠海马CA3区神经元形态学影响,以及慢性砷中毒对成年大鼠脑的神经毒性。方法:取3~4月龄SD大鼠60只,随机分为正常对照组(自由饮用蒸馏水)、低剂量组(As2O3按4mg/l的比例用蒸馏水配制,自由饮用)和高剂量组(As2O3按100mg/l的比例用蒸馏水配制,自由饮用)。每组20只,各组均用普通饲料喂养,连续喂养4月后取大鼠海马组织,常规石蜡切片尼式染色(Nissl),并利用神经元特异性烯醇化酶(NSE)抗体进行SABC法免疫组化染色,观察大鼠海马CA3区神经元的形态学改变,进行细胞形态学计数及测量NSE阳性反应产物的平均光密度。结果:砷中毒组神经元数量减少,细胞形态不规则,可见大量坏死的神经细胞,NSE免疫组化染色阳性细胞减少(P<0.01),阳性反应产物平均光密度降低(P<0.01)。结论:慢性砷中毒可以导致大鼠海马CA3区神经元结构损伤。
OBJECTIVE: To observe the effects of chronic arsenic poisoning on neuronal morphology in hippocampal CA3 region of adult rats and the neurotoxicity of chronic arsenic poisoning in adult rat brain by neuron-specific enolase (NSE) immunohistochemical staining. . Methods: Sixty Sprague-Dawley (SD) rats aged 3 ~ 4 months were randomly divided into normal control group (free drinking distilled water), low dose group (As2O3 treated with distilled water at a ratio of 4mg / l, free to drink) and high dose group According to the ratio of 100mg / l formulated with distilled water, free to drink). Twenty rats in each group were fed with normal diet. After continuous feeding for 4 months, hippocampal tissue of rats were taken out, Nissl was paraffin-embedded, and SABC method was performed using NSE antibody Immunohistochemical staining was used to observe the morphological changes of neurons in hippocampal CA3 region of rats. The cell morphology was counted and the average optical density of NSE-positive reaction products was measured. Results: The arsenic poisoning group had fewer neurons and irregular cell morphology, showing a large number of necrotic neurons. The number of NSE-positive cells decreased (P <0.01) and the average optical density of the positive reaction products decreased (P <0.01). Conclusion: Chronic arsenic poisoning can cause structural damage of neurons in hippocampal CA3 region of rats.
