目的探讨益胃汤延缓初老雌性大鼠卵巢机能衰老的机理。方法4~6月龄雌性SD大鼠为正常对照组;10~12月龄、阴道细胞学表现动情期延长的雌性SD大鼠作为初老大鼠模型,随机分为益胃汤高剂量组、益胃汤中剂量组、益胃汤低剂量组、己烯雌酚组、模型对照组。灌药4周后,取左侧卵巢用于检测卵巢Bcl-2、Bax、Caspase3、细胞色素C。结果与模型对照组比较,益胃汤可使卵巢Bcl-2表达增强,Bax表达减弱,Bcl-2/Bax比例增加(P<0.05),Caspase表达减少,细胞色素C减少,卵巢细胞凋亡减少,但达不到正常对照组水平(P>0.05),且益胃汤高、中、低剂量组在实验中呈现一定的量效关系。结论益胃汤通过抑制线粒体通路引起的级联反应抑制初老雌性大鼠卵巢细胞凋亡,这可能是其延缓卵巢机能衰老的机理之一。
Objective To investigate the mechanism of Yizhitang on slowing the ovarian function of young female rats. Methods Female Sprague-Dawley rats of 4 to 6 months old were normal control group. Female Sprague-Dawley rats, aged 10-12 months with prolonged estrous cycle of vaginal cytology, were randomly divided into Yisweitang high dose group, Stomach decoction medium dose group, Yiwei decoction low dose group, Diethylstilbestrol group, model control group. Four weeks after irrigation, the left ovary was taken for the detection of ovarian Bcl-2, Bax, Caspase3 and cytochrome C. Results Compared with the model control group, Yiwei decoction could enhance the expression of Bcl-2, decrease the expression of Bax, increase the ratio of Bcl-2 / Bax (P <0.05), decrease the expression of Caspase, decrease the cytochrome C and decrease the apoptosis of ovary , But it did not reach the level of the normal control group (P> 0.05). The Yisweitang high, medium and low dose groups showed a certain dose-effect relationship in the experiment. Conclusions Yiwei decoction can inhibit apoptosis of ovariectomized early-aged female rats by inhibiting the cascade reaction induced by mitochondrial pathway, which may be one of its mechanisms to delay ovarian function aging.
