目的 :观察硫丹 (ES)对培养的PC12 细胞增殖和形态学的影响及其诱导PC12 细胞凋亡作用。方法 :不同浓度MPP+ 和ES分别干预PC12 细胞 2 4h ,利用TUNEL染色、流式细胞仪、电镜检测观察其结果。结果 :① 1mmol·L-1MPP+ 组细胞数明显下降并有形态学改变 ;而 0 0 3mmol·L-1ES组细胞数就明显下降和有形态学改变 ,以 0 0 1mmol·L-1组最明显。②MPP+ 和ES均可诱导PC12 细胞凋亡 ,电镜下 0 0 6mmol·L-1ES组有较多凋亡细胞和少量坏死细胞。结论 :ES对PC12 细胞有毒性作用 ,其机制可能与细胞凋亡有关
Objective: To observe the effect of endosulfan (ES) on the proliferation and morphology of cultured PC12 cells and the induction of PC12 cell apoptosis. Methods: PC12 cells were treated with different concentrations of MPP + and ES for 24 hours respectively. TUNEL staining, flow cytometry and electron microscopy were used to observe the results. RESULTS: ① The number of cells in 1 mmol·L-1MPP + group decreased significantly and the morphological changes were observed. However, the number of cells in 0 0 3 mmol·L-1ES group was significantly decreased and the morphological changes were observed, the most obvious was in 0 0 1 mmol·L-1 group . ②MPP + and ES could induce apoptosis of PC12 cells. There were more apoptotic cells and a few necrotic cells in the 0 0 6mmol·L-1ES group under electron microscope. Conclusion: The toxic effect of ES on PC12 cells may be related to apoptosis
