目的:优选柚寄生总黄酮的提取工艺,并初步评价总黄酮体外抑制人肝癌细胞Hep3B的作用。方法:以芦丁为对照品测定柚寄生提取液的总黄酮含量,选取提取时间、提取温度、料液比及乙醇体积分数为影响因素,采用正交试验法优选柚寄生总黄酮的提取工艺条件,采用单因素试验考察提取次数;采用MTT法测定总黄酮对人肝癌细胞Hep3B的体外抑制作用。结果:优选的提取工艺为加30倍量60%乙醇于80℃提取2次,每次2 h;体外抗肿瘤活性试验结果表明,总黄酮质量浓度500 mg·L-1时对人肝癌细胞Hep3B的生长抑制率51.40%。结论:优选出的柚寄生总黄酮提取工艺条件稳定可行,柚寄生总黄酮有一定的体外抑制人肝癌细胞Hep3B的作用。
OBJECTIVE: To optimize the extraction process of total flavonoids from Citrus sinensis, and to evaluate the effect of total flavonoids on Hep3B cells in vitro. Methods: The content of total flavonoids in grapefruit parasitic extract was determined with rutin as the reference substance. The extraction time, extraction temperature, ratio of solid to liquid and ethanol volume fraction were selected as the influencing factors. The optimum extraction conditions of total flavonoids from grapefruit was determined by orthogonal test. Single factor experiments were used to examine the number of extractions; MTT assay was used to determine the inhibitory effect of total flavonoids on Hep3B cells. RESULTS: The optimal extraction process was extraction of 30 times 60% ethanol twice at 80°C for 2 h each time; the results of in vitro anti-tumor activity test showed that the total flavonoids concentration 500 mg·L-1 in human hepatoma cells Hep3B. The growth inhibition rate was 51.40%. Conclusion: The optimized extraction conditions of total flavonoids from Citrus sinensis are stable and feasible. The total flavonoids from Citrus sinensis can inhibit the function of Hep3B cells in vitro.
