目的 了解肝细胞肝癌患者血清和组织细胞间粘附分子 1(ICAM 1)及其mRNA的表达 ,明确血清可溶性ICAM 1(sICAM 1)的来源。方法 应用酶联免疫吸附试验 (ELISA)分析、免疫组织化学及逆转录 聚合酶链式反应 (RT PCR)等技术 ,测定 15 1例肝细胞肝癌 (HCC)血清、组织ICAM 1及其mRNA的表达 ,并与甲胎蛋白 (AFP)进行同步分析。结果 15 1例HCCsICAM 1明显高于各对照组 ,73 %的患者sICAM 1>10 0 0 μg/L。肝癌组织中ICAM 1的表达阳性率为94.2 % ,AFP为 62 .9%。ICAM 1mRNA的表达与肝细胞的恶变与否及患者血清sICAM 1的水平密切相关 ,它在癌组织及癌旁组织中的表达率分别为 65 .1%和 3 0 .2 % ,正常肝组织均无表达。结论 HCC患者血清sICAM 1的高水平与其蛋白分子在组织中的过度表达一致 ,并受其相关基因的调控 ,肝细胞恶变则是sICAM 1表达的病理学基础。测定sICAM 1对监测HCC及其术后复发具有一定的临床意义
Objective To understand the expression of intercellular adhesion molecule-1 (ICAM 1) and its mRNA in serum and tissue of patients with hepatocellular carcinoma (HCC) and to determine the origin of soluble ICAM 1 (sICAM 1). Methods The expression of ICAM - 1 and its mRNA in serum and tissue of 15 cases of hepatocellular carcinoma (HCC) were determined by enzyme linked immunosorbent assay (ELISA), immunohistochemistry and reverse transcription polymerase chain reaction (RT PCR) , And simultaneously analyzed with AFP (AFP). Results 15 cases of HCCs ICAM 1 was significantly higher than the control group, 73% of patients with sICAM 1> 10 0 0 μg / L. The positive rate of ICAM 1 expression in hepatocellular carcinoma was 94.2% and AFP was 62.9%. The expression of ICAM 1 mRNA was closely related to the malignancy of hepatocytes and the level of serum sICAM 1. The expression rates of ICAM 1 mRNA in cancer tissues and adjacent tissues were 65.1% and 32.2% No expression. Conclusion The high level of serum sICAM 1 in HCC patients is consistent with the overexpression of its protein in the tissues and is regulated by its related genes. The malignant transformation of hepatocytes is the pathological basis of sICAM 1 expression. Determination of sICAM 1 has certain clinical significance for monitoring HCC and its recurrence
