目的:观察系膜细胞是否影响补肾药物对成骨细胞的作用。方法:空白组大鼠灌服蒸馏水,药物组大鼠以熟地鳖甲1:1配比制成浸膏,3.3g浸膏/kg体质量/d灌服,3d后取血离心,获得无药血清和熟地鳖甲含药血清。无药血清、熟地黄鳖甲含药血清及肾系膜细胞处理的熟地鳖甲含药血清作用于成骨细胞3d,MTT法观察成骨细胞增殖率,实时荧光定量PCR检测成骨细胞Ⅰ型胶原(collagenⅠ)和核心结合因子α1(cbfα1)基因表达。结果:熟地黄、鳖甲含药血清可促进成骨细胞增殖,经肾系膜细胞处理后的含药血清进一步促进细胞增殖(P<0.05),荧光定量PCR显示含药血清组成骨细胞collagenⅠ和cbfα1基因表达量明显上调(P<0.05),经肾系膜细胞处理后的含药血清2种基因表达量进一步增加(P<0.05)。结论:肾脏固有细胞——肾系膜细胞可在补肾药物基础上进一步促进成骨细胞的增殖和成骨相关基因的表达。
Objective: To observe whether mesangial cells affect the effect of kidney-tonifying drugs on osteoblasts. Methods: The rats in the blank group were drenched with distilled water. The rats in the drug group were treated with the ratio of 1: 1 of the turtle, and 3.3g extract / kg body weight / d was given orally. After 3d, the blood was centrifuged to obtain the drug Serum and Rehmannia turtle containing serum. No drug serum, Radix Rehmanniae Preparata containing serum and mesangial cells treated Rehmannia turtle contains serum on osteoblasts 3d, MTT assay osteoblast proliferation rate, real-time fluorescent quantitative PCR detection of osteoblast type Ⅰ Collagen Ⅰ and corebinding factor α1 (cbfα1) gene expression. Results: Radix Rehmanniae Preparata and turtle shell containing drug serum could promote the proliferation of osteoblasts. The drug-containing serum treated by mesangial cells further promoted cell proliferation (P <0.05). Fluorescent quantitative PCR showed that the drug-containing serum contained collagen Ⅰ and The expression of cbfα1 gene was significantly up-regulated (P <0.05). The expression of two genes in the serum treated with mesangial cells further increased (P <0.05). CONCLUSION: Renal intrinsic cell mesangial cells can further promote the proliferation of osteoblasts and the expression of osteogenesis-related genes on the basis of kidney-invigorating drugs.
