目的观察肝硬化大鼠胃动力的变化,并探讨肝硬化胃肠动力障碍的相关机制。方法20只Wistar大鼠随机分为肝硬化模型组和对照组,采用四氯化碳溶剂大腿根部皮下注射法制作肝硬化模型,液相胃排空检测法测定胃排空,RT-PCR法检测大鼠胃窦平滑肌细胞色素氧化酶Ⅱ基因(COⅡgene)的变化,免疫组化染色观察胃窦C-kit阳性Cajal间质细胞的变化,并用透射电镜观察胃窦Ca-jal间质细胞的超微结构改变。结果与对照组比较,肝硬化组大鼠排空率明显下降,胃窦平滑肌COⅡmRNA的表达显著低于对照组(P<0.01);胃窦C-kit阳性Cajal间质细胞明显减少(P<0.01),透射电镜观察显示Cajal间质细胞与其他细胞间连接减少,线粒体肿胀,细胞器减少,胞浆内空泡形成。结论肝硬化大鼠胃排空明显下降,胃窦平滑肌COⅡmRNA表达及Cajal间质细胞发生明显变化,胃窦平滑肌COⅡmRNA表达下降、Cajal间质细胞减少及其超微结构改变可能与肝硬化大鼠胃动力障碍有关。
Objective To observe the changes of gastric motility in cirrhotic rats and to explore the related mechanisms of gastrointestinal motility disorders in cirrhosis. Methods Twenty Wistar rats were randomly divided into liver cirrhosis model group and control group. Liver cirrhosis model was made by subcutaneous injection of carbon tetrachloride solvent thigh. Gastric emptying was measured by liquid gastric emptying. RT-PCR The changes of CO Ⅱ gene in rat gastric antral smooth muscle cells were observed. The changes of C-kit positive Cajal interstitial cells in gastric antrum were observed by immunohistochemistry. The ultrastructure of Ca-jal interstitial cells in gastric antrum Structural change. Results Compared with the control group, the emptying rate of cirrhotic rats decreased significantly, the expression of COⅡmRNA in gastric smooth muscle was significantly lower than that of the control group (P <0.01); the C-kit positive Cajal interstitial cells in gastric antrum significantly decreased (P <0.01 ), Transmission electron microscopy showed Cajal interstitial cells and other cells decreased connectivity, mitochondria swelling, organelles decreased cytoplasmic vacuoles formation. Conclusions The gastric emptying in cirrhotic rats is obviously decreased, the expression of COⅡmRNA and the expression of Cajal interstitial cells in gastric antrum significantly decrease, the expression of COⅡmRNA in gastric antral smooth muscle decreases, the expression of Cajal interstitial cells and ultrastructure changes may be related to the gastric Power related.
