目的以云南省不同地区云南重楼及多芽品系为材料,采用UPLC-ELSD法同时测定7种甾体皂苷量并建立高黎贡山地区样品UPLC指纹图谱,评价其质量。方法采用UPLC-ELSD法,ACQUITY UPLC BEH C18(50 mm×2.1 mm,1.7μm)色谱柱,体积流量0.2 m L/min,漂移管温度55℃,氮气压力275.8 k Pa,增益500,以乙腈-水为流动相梯度洗脱。结果样品在23 min内能完全被分离,方法学考察均符合要求;23份样品中19份符合《中国药典》2015年版要求,《中国药典》项下4个皂苷平均质量分数为1.42%,其中高黎贡山地区的样品平均质量分数高达1.660%;10批次高黎贡山地区云南重楼(多芽品系)样品间相似度小于0.9,标定了8个共有特征峰,并对4个特征峰进行了归属;样品中7个皂苷的种类和量经PCA分析和系统聚类分析均不能明显分类。结论该方法简便、快速、灵敏度高,可用于云南重楼的质量快速评价;云南重楼及其多芽品系在化学成分种类和量上并无明显区别;高黎贡山地区的云南重楼(多芽品系)是一个值得推广栽培和深入研究的品种。
OBJECTIVE To determine the amount of seven steroidal saponins by UPLC-ELSD and to establish the UPLC fingerprinting in Gaoligongshan region by using the re-building and the multi-buds in different regions of Yunnan Province. Conclusions The method is simple, rapid and sensitive and can be used for the rapid assessment of the quality of re-building in Yunnan. There are no significant differences in the chemical composition types and quantities between Rei-Lou and its multi-bud lines in Yunnan.
