目的观察弥漫性轴索损伤(diffuse axonal injury,DAI)后大鼠海马CA1区锥体细胞自发放电的特征,揭示DAI对大鼠海马CAl区锥体神经元兴奋性的影响。方法雄性健康成年SD大鼠30只,随机分为正常对照组及DAI后6h组、12h组、24h组、72h组。采用玻璃微电极细胞外记录的方法记录海马神经元的放电活动,每只动物记录2个单位放电。分析比较各组各个单位放电的放电类型、放电频率等差异。结果 1DAI后各组与正常组大鼠海马CA1区锥体细胞观察到3种类型放电,即不规则放电、规则放电和簇式放电。在放电类型上各组差异无统计学意义。2比较各组的12个单位放电的平均放电频率、平均ISI可得出DAI后12h组的平均放电频率低于其他组,差异有统计学意义;其他组比较差异则无统计学意义。3比较各组簇式放电的簇内ISI:DAI后12h组最小,其次是24h组、72h组,6h组与正常组最大且差异无统计学意义。结论 DAI对大鼠海马CAl区锥体神经元兴奋性具有明显影响,并且呈不同放电类型的动态变化。
Objective To observe the characteristics of spontaneous discharge in rat hippocampal CA1 pyramidal cells after diffuse axonal injury (DAI), and to reveal the effect of DAI on the excitability of pyramidal neurons in hippocampal CA1 region of rats. Methods Thirty male adult SD rats were randomly divided into normal control group and 6h, 12h, 24h and 72h after DAI. The extracellular recordings of glass microelectrodes were used to record the discharge activity of hippocampal neurons. Each animal recorded 2 units of discharge. Analysis and comparison of discharge discharge types, discharge frequency and other differences among various units in each group. Results Three types of discharges were observed in hippocampal CA1 pyramidal cells of rats in each group and normal group after 1 DAI, namely irregular discharge, regular discharge and cluster discharge. There was no significant difference in discharge types between the groups. 2 The average discharge frequency of 12 units in each group was compared. The average ISI showed that the average discharge frequency of 12h after DAI was lower than other groups, the difference was statistically significant; the other groups had no statistical significance. The cluster ISI DAI was the smallest at 12h, followed by the 24h, 72h and 6h groups, and the difference was not statistically significant. Conclusion DAI has a significant effect on the excitability of pyramidal neurons in hippocampal CA1 region of rats, and shows dynamic changes in different discharge types.
