目的:建立RP-HPLC法同时测定不同药用部位、不同产地、不同季节的旱芹中绿原酸和咖啡酸的含量。方法:采用Kromasil C18柱(5μm,250mm×4.6mm),流动相为甲醇-0.05%磷酸水溶液梯度洗脱,流速1.0mL.min-1,检测波长327nm,柱温35℃。结果:绿原酸和咖啡酸进样浓度分别在3.71~92.8μg.mL-1(r=0.9999)和0.148~3.70μg.mL-1(r=0.9997)范围内与峰面积呈良好的线性关系,平均加样回收率(n=9)分别为100.3%和100.2%。结论:分析方法快速、准确,重现性好,可用于控制旱芹的质量。
OBJECTIVE: To establish an RP-HPLC method for simultaneous determination of chlorogenic acid and caffeic acid in celery with different medicinal parts, different producing areas and different seasons. Methods: Kromasil C18 column (5μm, 250mm × 4.6mm) was used. The mobile phase consisted of a gradient of methanol-0.05% phosphoric acid. The flow rate was 1.0mL.min-1. The detection wavelength was 327nm and the column temperature was 35 ℃. RESULTS: The concentrations of chlorogenic acid and caffeic acid were linearly correlated with the peak area in the range of 3.71-92.8 μg.mL-1 (r = 0.9999) and 0.148-3.70 μg.mL-1 (r = 0.9997), respectively , The average recovery rate (n = 9) was 100.3% and 100.2% respectively. Conclusion: The analytical method is rapid, accurate and reproducible. It can be used to control the quality of celery.
