为观察氧化型低密度脂蛋白对内皮细胞一氧化氮生成的影响,采用体外细胞培养方法将含不同浓度氧化型低密度脂蛋白的DMEM 培养液( 氧化型低密度脂蛋白终浓度分别为0 mgL、50 mgL 、100 mgL 、200 mgL),与内皮细胞共同孵育(37 ℃、5% CO2)24 h 后,分别测上清液中乳酸脱氢酶活性和一氧化氮含量及细胞中一氧化氮合酶活性,并用细胞化学染色法记数各组细胞一氧化氮合酶阳性染色细胞数。结果发现,随着培养液中氧化型低密度脂蛋白浓度升高,上清液中乳酸脱氢酶活性增加和一氧化氮含量下降,细胞中一氧化氮合酶活性和一氧化氮合酶阳性染色细胞数下降,可见,氧化型低密度脂蛋白可损伤血管内皮细胞,降低细胞中一氧化氮合酶活性,抑制一氧化氮产生,这可能是氧化型低密度脂蛋白致动脉粥样硬化的原因之一。
In order to observe the effect of oxidized low-density lipoprotein on nitric oxide production in endothelial cells, in vitro culture of DMEM containing different concentrations of oxidized low density lipoprotein (final concentrations of oxidized low density lipoprotein were 0 mg L, 50 mg L, 100 mg L, 200 mg L) were incubated with endothelial cells (37 ℃, 5% CO2) for 24 h.
