目的:分析不同预后小肠间质瘤病人组织蛋白质质点,寻找判断小肠间质瘤预后的候选蛋白质。方法:预后不良组和预后良好组小肠间质瘤组织各5例,经双向凝胶电泳分离总蛋白质,运用图像分析软件分析,对差异蛋白质点采用基质辅助激光解析电离飞行时间质谱仪鉴定部分差异蛋白质点。结果:比较蛋白质组学共鉴定出蛋白质28个(预后不良组中9个蛋白质表达上调,19个蛋白质下调)。预后不良组小肠间质瘤组织中微管不稳定蛋白(stathmin)、血清结合珠蛋白(haptoglobin 2,HP-2)和微管蛋白表达明显升高。结论:不同预后小肠间质瘤组织的蛋白质谱存在差异,stathmin和HP-2及微管蛋白可能成为判断小肠间质瘤病人预后的3种因子,为今后小肠间质瘤病人的临床预后判断提供依据。
OBJECTIVE: To analyze the tissue protein spots of patients with different prognosis of small intestinal stromal tumors and to search for candidate proteins for judging the prognosis of small intestinal stromal tumors. Methods: Five patients with poor prognosis group and good prognosis group were divided into two groups according to the two-dimensional gel electrophoresis. Total protein was separated by two-dimensional gel electrophoresis and analyzed by image analysis software. The difference of protein spots was identified by matrix-assisted laser desorption / ionization time of flight mass spectrometry Protein spots. RESULTS: A total of 28 proteins were identified by comparative proteomics (9 proteins in the poor prognosis group were up-regulated and 19 proteins were down-regulated). In the poor prognosis group, the expression of stathmin, HP-2 and tubulin were significantly increased in small intestinal stromal tumors. CONCLUSIONS: There are differences in the protein profiles of different prognosis of small intestinal stromal tumors. Stathmin, HP-2 and tubulin may be the three prognostic factors for evaluating the prognosis of small intestinal stromal tumors and provide a basis for the future clinical judgment of small intestinal stromal tumors in accordance with.
