目的观察通肺平哮汤对卵蛋白致敏哮喘豚鼠小气道SP阳性纤维表达的影响,探讨通肺平哮汤治疗哮喘的机制。方法 40只健康雄性Hartley系豚鼠随机分为空白对照组、模型组、地塞米松组和通肺平哮汤组。用卵白蛋白致敏,激发构建哮喘豚鼠模型,用免疫组化方法检测SP阳性纤维在豚鼠肺内的表达。结果 通肺平哮汤组和地塞米松组SP阳性纤维则较模型组显著减少,通过模型组肺组织HE切片电镜观察证实该组气道壁及肺间质中嗜酸性粒细胞大量浸润,气道平滑肌层增厚,管腔狭窄,毛细血管内皮肿胀。结论通肺平哮汤可抑制气道炎症反应,且加速SP降解,抑制神经源性炎症而发挥治疗哮喘的作用。
Objective To observe the effects of Tongfei Pingxiao decoction on the expression of small airway SP positive fibers in asthmatic guinea pigs induced by ovalbumin and to explore the mechanism of Tonglupingxiao decoction in treating asthma. Methods Forty healthy male Hartley guinea pigs were randomly divided into blank control group, model group, dexamethasone group and Tongfei Pingxiao decoction group. Sensitized with ovalbumin, the model of asthmatic guinea pigs was established and the expression of SP positive fibers in guinea pig lungs was detected by immunohistochemistry. Results Compared with the model group, the SP-positive fibers in Tongfei Pingtong Decoction group and dexamethasone group were significantly reduced. The eosinophilic granulocyte infiltration in the airway wall and interstitium was confirmed by HE section electron microscopy in the model group Road smooth muscle thickening, stenosis, capillary endothelial swelling. Conclusion Tongfang Pingxiao Decoction can inhibit the airway inflammatory response, accelerate the degradation of SP, and inhibit the neurogenic inflammation and play a role in the treatment of asthma.
