目的:研究三七总皂苷(PNS)对快速老化SAMP8小鼠大脑α分泌酶含量和活性的影响。方法:采用快速老化痴呆模型小鼠SAMP8,将其随机分为模型组、PNS高剂量组(200 mg/kg)、PNS低剂量组(100 mg/kg)和石杉碱甲组(0.3 mg/kg),8只/组,对照组给予给药组相同容积的双蒸水灌胃。每天灌胃1次,连续给药2个月。分别采用western blot和直接免疫荧光法检测三七总皂苷对快速老化SAMP8小鼠大脑α分泌酶(ADAM 9、AD-AM10、ADAM17)的影响。结果:western blot结果显示PNS高、低剂量组ADAM10和ADAM17蛋白表达均低于模型组(P<0.05),而ADAM9蛋白表达量均高于模型组(P<0.05);石杉碱甲组ADAM17、ADAM9蛋白表达量均高于模型组(P<0.05),而ADAM10蛋白表达则低于模型组(P<0.05)。直接免疫荧光法结果显示PNS高、低剂量组和石杉碱甲的RFU值均高于模型组(P<0.01)。结论:三七总皂苷可以通过提高α分泌酶中的ADAM9的含量,石杉碱甲则可能通过提高ADAM9和ADAM17的含量,从而提高SAMP8小鼠大脑α-Secretase的活性。
Objective: To study the effects of Panax Notoginseng Saponins (PNS) on the content and activity of α-secretase in the brain of rapidly aged SAMP8 mice. Methods: SAMP8 was used in the model mice of fast-aging dementia. The rats were randomly divided into model group, PNS high dose group (200 mg / kg), PNS low dose group (100 mg / kg) and Huperzine A group (0.3 mg / kg), 8 / group, the control group was administered the same volume of double distilled water gavage. Gavage 1 day, continuous administration for 2 months. The effects of Panax notoginseng saponins on α-secretase (ADAM 9, AD-AM10, ADAM17) in the brain of rapidly aged SAMP8 mice were detected by western blot and direct immunofluorescence respectively. Results: Western blot results showed that the protein expressions of ADAM10 and ADAM17 were lower in PNS high and low dose groups than those in model group (P <0.05), while the expression of ADAM9 protein was higher in model group (P <0.05) , And the expression of ADAM9 protein in model group were higher than those in model group (P <0.05), while the expression of ADAM10 protein was lower than model group (P <0.05). The results of direct immunofluorescence showed that the RFU values of PNS high, low dose and huperzine A were higher than those of model group (P <0.01). Conclusion: Panax notoginseng saponins can increase the content of ADAM9 in α-secretase. Huperzine A may increase the activity of α-Secretase in brain of SAMP8 mice by increasing the contents of ADAM9 and ADAM17.
