Objective To optimize the experiment conditions for ultrasonic extraction of total saponin from Codonpsis pilosula.Method The orthogonal test design was adopted to optimize the factors of the extraction process of total saponin in Codonpsis pilousla,and the colorimetric method was used to determine the content of total saponin.The new method was compared with the traditional extraction methods.Result The optimal ultrasonic extraction conditions were as follows: methanol was used as the extracting solvent,the ratio of solid to liquid was 1∶20 (W/V,g/mL) and ultrasonic extraction was conducted three times with 30 minutes each time.Under these conditions,the total saponin extracted from Codonpsis pilosula was 65.48 mg/g.Conclusion This method is simple with stable conditions,and it improves utilization of medical material,which can be used to extract total saponin from Codonpsis pilosula.
