目的:胚胎干细胞是具有全能性及无限制自我更新与分化能力的一类特殊细胞群体,通过对胚胎干细胞诱导分化研究的回顾和总结,期望能给胚胎干细胞的临床应用提供理论参考。资料来源:应用计算机检索Highwirepress以及Medline数据库2000/2004期间的相关文章,检索词为“Embryonicstemcell,differentiation,induce,同时计算机检索万方数据库2000/2004期间的相关文章,检索词为“胚胎干细胞”,限定文章类型为中文。资料选择:对资料进行初审,选取与诱导分化有关的文章,纳入标准为研究原著。资料提炼:选取与诱导分化有关的文章共369篇(其中外文364篇)。通过阅读相关文献的摘要,从中选取15篇较全面介绍胚胎干细胞的诱导分化以及应用的文章。其中,全面介绍胚胎干细胞各种细胞模型,神经细胞模型,心肌细胞模型,上皮细胞模型,造血细胞模型。资料综合:7篇文献较为全面的介绍胚胎干细胞可以诱导分化为神经细胞、心肌细胞、上皮细胞以及造血细胞等细胞模型,从以上文献以及单独介绍各种细胞模型的文章中分别对各种细胞模型的研究概况进行归纳整理。结论:胚胎干细胞经过定向诱导分化后可获得神经细胞、心肌细胞、上皮细胞以及造血细胞等细胞模型,这些模型可以用于细胞发育分化的分子机制,并对药物筛选和临床细胞治疗提供良好应用前景。
OBJECTIVE: Embryonic stem cells (ESCs) are a kind of special cell population with pluripotency and unlimited self-renewal and differentiation ability. Data selection: Preliminary review of the data, selection of articles related to the induction of differentiation, inclusion criteria for the study of the original data extraction: 369 articles (including 364 foreign language articles) were selected for the induction of differentiation. 15 articles were selected to introduce the induction and differentiation of embryonic stem cells and their applications in detail.Among them, we introduced various cell models, neural cell model, cardiomyocyte model, epithelial cell model and hematopoietic cell model of embryonic stem cells in detail.
