分别以MTT法和图像分析法检测了3种感染根管内厌氧菌纯化荚膜对成纤维细胞L_(929)的细胞毒性。实验结果显示:3种纯化荚膜中以牙龈卟啉菌荚膜毒性最强,中间型普里沃氏菌次之,牙髓卟啉菌荚膜毒性不明显,纯化荚膜细胞毒性对MTT检测法不敏感。3种纯化荚膜对成纤维细胞的毒性作用与内毒素有所不同,主要是使细胞和细胞核面积增大,且以细胞面积增大为著。
MTT assay and image analysis were used to detect the cytotoxicity of anaerobic bacteria-purified capsule on L929 cells in three kinds of infected root canals. The experimental results showed that the three purified capsulas were the most toxic to the capsular strain of Porphyromonas gingivalis, followed by Prunella intermedium, the capsule toxicity of Porphyromonas gingivalis was not obvious, and the cytotoxicity of purified capsular membrane was sensitive to MTT assay Law is not sensitive. The three purified capsular fibroblasts toxic effects and endotoxin are different, mainly to make the cell and nucleus area increased, and the cell area for the increase.
