目的探讨高脂膳食对小鼠以及槲皮素对高脂膳食小鼠学习记忆能力的影响。方法 60只昆明种雄性小鼠,随机分为对照组(正常膳食),高脂膳食组(含18%猪油),槲皮素低剂量组(高脂膳食+0.005%槲皮素)和槲皮素高剂量组(高脂膳食+0.01%槲皮素)。12w后进行Morris水迷宫测试小鼠的学习记忆能力,检测小鼠全血和脑组织的自由基和氧化应激指标并用荧光定量PCR检测海马脑源性神经生长因子(BDNF)的相对表达量。结果高脂膳食使小鼠的学习记忆能力显著下降(P<0.05),脑组织抗氧化能力显著降低(P<0.01),血浆和脑组织丙二醛(MDA)、自由基水平显著升高(P<0.05),海马BDNF的相对表达显著降低(P<0.05)。添加槲皮素后脑部的氧化还原状况得到显著改善(P<0.05)槲皮素高剂量组BDNF基因的相对表达量显著升高(P<0.05)。结论高脂膳食可导致慢性氧化应激,损伤小鼠的学习记忆能力。槲皮素可改善脑部氧化还原状态,增加BDNF的表达,从而改善小鼠的学习和记忆能力。[营养学报,2012,34(4):353-357]
Objective To investigate the effects of high fat diet on learning and memory ability of mice and quercetin on high fat diet mice. Methods Sixty Kunming male mice were randomly divided into control group (normal diet), high fat diet group (containing 18% lard), low dose quercetin group (high fat diet + 0.005% quercetin) and quercetin High-dose dermatin group (high-fat diet + 0.01% quercetin). After 12 weeks, Morris water maze test was used to study the learning and memory abilities of mice. Free radicals and oxidative stress indexes were detected in whole blood and brain of mice. The relative expression of BDNF in hippocampus was detected by real-time PCR. Results The high fat diet decreased the learning and memory abilities of mice (P <0.05), the antioxidant capacity of brain tissue decreased significantly (P <0.01), the levels of malondialdehyde (MDA) and free radicals in plasma and brain tissue increased significantly P <0.05). The relative expression of BDNF in hippocampus decreased significantly (P <0.05). After addition of quercetin, the brain redox status was significantly improved (P <0.05). The relative expression of BDNF gene in quercetin high dose group was significantly increased (P <0.05). Conclusion High-fat diet can cause chronic oxidative stress and impair the learning and memory ability of mice. Quercetin can improve brain redox status, increase BDNF expression, thereby improving mice learning and memory. [Journal of Nutrition, 2012,34 (4): 353-357]
