目的 18F-FDG PET/CT显像评价肝癌H22模型及三萜类中药成分熊果酸(UA)对移植性肝癌(H22)细胞体内生长的抑制作用。方法昆明小鼠20只,雌雄各半,体重(20±2)g,右前腋皮下接种1×107/0.2 ml小鼠肝癌H22细胞,左前腋皮下接种等量生理盐水作为对照。建立实体瘤模型并进行PET/CT检查评价实体瘤模型后,随机分两组,对照组和熊果酸组(UA组),每组10只,UA组每只按4.536 mg/kg给予体积0.2 ml的UA,每天1次,连续10 d;对照组给相同容积的生理盐水。给药结束后再次行18F-FDG PET/CT显像。显像后处死小鼠,采用HE染色法检测形态学改变。结果 (1)20只小鼠治疗前右前腋皮下肿瘤接种部位放射性摄取/同侧肺(T右1/NT右1)为2.833±0.18,左前腋皮下接种等量生理盐水区放射性摄取/同侧肺(T左1/NT左1)为1.078±0.27,T右1/NT右1与T左1/NT左1有统计学差异(t=15.55,P=0.0000)。(2)对照组与UA组右前腋皮下接种肿瘤部位放射性摄取/同侧肺(T右2/NT右2与T右3/NT右3)分别为2.824±0.224及1.534±0.429,二者有统计学差异(t=11.85,P=0.0001)。左前腋皮下同等体积生理盐水接种放射性摄取/同侧肺(T左2/NT左2与T左3/NT左3)分别为1.078±0.33及1.076±0.34,二者无统计学差异(t=2.121,P=0.078)。结论18F-FDG PET/CT显像是评价肝癌H22模型及熊果酸抑制肝癌效果的准确方法。
Objective To evaluate the inhibitory effect of 18F-FDG PET / CT imaging in H22 hepatocarcinoma and the effect of UA on the growth of human hepatocellular carcinoma (H22) cells. Methods Twenty Kunming mice, male and female, weighing 20 ± 2 g and 1 × 107 / 0.2 ml H22 cells were inoculated subcutaneously into the right anterior arches. The rats were injected subcutaneously with normal saline as control. The solid tumor model was established and PET / CT examination was performed to evaluate the solid tumor model. Two groups were randomly divided into control group and ursolic acid group (UA group), 10 rats in each group. The volume of UA group was 4.5 ml of UA once a day for 10 days; the control group to the same volume of saline. 18F-FDG PET / CT imaging was performed again after the end of administration. After imaging, mice were sacrificed and morphological changes were detected by HE staining. Results (1) Radioactive uptake / ipsilateral lung (T right 1 / NT right 1) in the right anterior subaxillary tumor in 20 mice before treatment was 2.833 ± 0.18, and the same dose Lung (T left 1 / NT left 1) was 1.078 ± 0.27, T right 1 / NT right 1 and T left 1 / NT left 1 were statistically different (t = 15.55, P = 0.0000). (2) The radioactive uptake / ipsilateral lung (T right 2 / NT right 2 and T right 3 / NT right 3) subcutaneously inoculated subcutaneously into the right forearm in the control and UA groups were 2.824 ± 0.224 and 1.534 ± 0.429, respectively Statistical difference (t = 11.85, P = 0.0001). There was no significant difference between the two groups (0.78 ± 0.33 and 1.076 ± 0.34, respectively) for radioactive uptake / ipsilateral lung (T left 2 / NT left 2 and T left 3 / NT left 3) 2.121, P = 0.078). Conclusion 18F-FDG PET / CT imaging is an accurate method to evaluate H22 model of liver cancer and ursolic acid inhibit liver cancer.
