目的:探讨阿霉素肾病(AN)大鼠肾组织匀浆超氧化物岐化酶(SOD)和丙二醛(MDA)水平及其临床意义。方法:一次性尾静脉注射阿霉素法制备AN大鼠模型,邻苯三酚自氧化法测定肾组织匀浆超氧化物岐化酶(SOD)活力,硫代巴比妥酸(TBA)法测定肾组织匀浆液丙二醛(MDA)水平。结果:AN大鼠尿中蛋白排泄量明显增加,电镜下可见肾小球上皮细胞足突融合,内皮细胞增生,严重者肾小球基膜增厚,模糊不清,内皮细胞呈连拱状。AN组大鼠血清TP和Alb水平低于对照组,而TG、TC、HDL、LDL、ApoA和ApoB水平则高于对照组,其肾组织匀浆MDA值(2.85±0.37)升高,SOD值(2.14±0.74)下降,与对照组比较,差异显著。结论:AN大鼠存在脂质过氧化,脂质代谢异常与其病理改变有关。
Objective: To investigate the levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in renal tissue of adriamycin nephropathy rats and its clinical significance. Methods: A rat model of AN was injected with doxorubicin through tail vein injection. The activity of superoxide dismutase (SOD) and the activity of thiobarbituric acid (TBA) in renal tissue homogenate were determined by pyrogallol autoxidation method. Malondialdehyde (MDA) level in renal homogenate was measured. Results: The protein excretion of urinary excretion was significantly increased in AN rats. Under electron microscopy, glomerular epithelial cell foot process fusion and endothelial cell proliferation were observed. Glomerular basement membrane was thickened, blurred and endothelial cells were even arched. The level of serum TP and Alb in AN group was lower than that in control group, while the levels of TG, TC, HDL, LDL, ApoA and ApoB were higher than those in control group. The MDA value in renal tissue homogenate (2.85 ± 0.37) (2.14 ± 0.74), compared with the control group, the difference was significant. Conclusion: There is lipid peroxidation in AN rats. The abnormal lipid metabolism is related to the pathological changes.
