目的探索HMBA及其衍生物HMBA-Ⅰ和HMBA-Ⅱ对人红白血病细胞系K562的诱导分化作用。方法细胞生长曲线绘制、集落形成试验,检测HMBA及其衍生物对细胞生长的影响,以Westernblot检测p21、c-fos蛋白表达,以联苯胺染色显示血红蛋白。结果HMBA-Ⅰ,HMBA-Ⅱ呈现对K562细胞生长有不同程度的抑制作用,基因表达产物血红蛋白联苯胺染色呈阳性反应,p21、c-fos蛋白表达有不同程度的增高。结论HMBA-Ⅰ,HMBA-Ⅱ对K562有一定的诱导分化活性,作用机制可能与p21、c-fos表达增高有关。
Objective To explore the differentiation of human erythroleukemia cell line K562 induced by HMBA and its derivatives HMBA-I and HMBA-II. Methods Cell growth curves and colony formation assays were performed to examine the effects of HMBA and its derivatives on cell growth. Western blot was used to detect the expression of p21 and c-fos proteins, and benzidine staining was used to show hemoglobin. Results HMBA-I and HMBA-II showed different degrees of inhibition on the growth of K562 cells. The gene expression product was positive for hemoglobin benzidine staining, and the expression of p21 and c-fos proteins increased to varying degrees. Conclusion HMBA-I and HMBA-II have a certain induction and differentiation activity on K562, and the mechanism may be related to the increased expression of p21 and c-fos.
