目的测定肌萎灵冻干粉中淫羊藿苷和朝藿定A、B、C 4种黄酮类成分的血浆蛋白结合率。方法采用平衡透析法结合高效液相色谱法,测定肌萎灵冻干粉中4种黄酮类成分在透析袋内血浆中的质量浓度与透析袋外缓冲液中的质量浓度,计算血浆蛋白结合率。结果肌萎灵冻干粉血浆浓度分别为20、10和5 g.L-1时,肌萎灵冻干粉中淫羊藿苷和朝藿定A、B、C与人血浆的蛋白结合率分别为(67.7±s 1.1)%、(73.4±0.4)%、(70.0±1.5)%;(56.3±2.4)%、(62.2±1.4)%、(62.0±1.4)%;(56.5±1.0)%、(59.7±1.5)%、(57.3±2.7)%;(62.4±1.4)%、(67.7±0.8)%、(66.8±1.8)%。结论肌萎灵冻干粉中4种黄酮类成分与人血浆均属中度结合,且蛋白结合率与血药浓度无明显依赖性。
Objective To determine the plasma protein binding of flavonoids A, B and C of icariin and epimedin in the freeze-dried powder of wispinger. Methods The dialyzed bag was used to determine the plasma protein binding rate of the four flavonoids in the lyophilized bag by HPLC with dialysis method and high performance liquid chromatography . Results The results showed that the protein binding rates of icariin and epimedin A, B, C to human plasma in muscle wilt freeze-dried powder were 20, 10 and 5 gL-1, respectively (67.2 ± 1.1)%, (73.4 ± 0.4)%, (70.0 ± 1.5)%; (56.3 ± 2.4)%, (62.2 ± 1.4)%, (62.0 ± 1.4)%; (59.7 ± 1.5)%, (57.3 ± 2.7)%; (62.4 ± 1.4)%, (67.7 ± 0.8)%, (66.8 ± 1.8)% respectively. Conclusion The four flavonoids in the freeze-dried muscle powder are moderately bound to human plasma, and the protein binding rate has no obvious dependence on the plasma concentration.
