本实验旨在探究普安银鲫(Carassius auratus gibelio)早期发育过程中氧化损伤程度和总抗氧化能力的变化,及添加维生素C(Vc)后对它们的影响。采用生化方法测定了普安银鲫早期发育过程中丙二醛(MDA)含量、蛋白质羰基(PCA)含量、乳酸脱氢酶(LDH)的活性和总抗氧化能力(T-AOC)。(1)30 mg/L的Vc溶液中仔鱼孵化率和成活率最高,分别高出对照组(Vc浓度为0 mg/L)16.2%和17.8%;(2)普安银鲫早期发育过程中丙二醛、蛋白质羰基含量及乳酸脱氢酶活性均呈“升高-降低”的趋势,均在原肠中期达到最高,总抗氧化能力逐渐增加;(3)30 mg/L的Vc组中丙二醛、蛋白质羰基含量及乳酸脱氢酶的活性显著低于对照组(P<0.05),总抗氧化能力显著高于对照组(P<0.05)。结果表明,(1)普安银鲫胚胎期受到的氧化损伤比胚后严重;(2)适宜浓度的维生素C能提高胚胎及仔鱼的抗氧化能力,有效降低胚胎及仔鱼受到的氧化损伤,提高其孵化率及存活率。
The purpose of this study was to investigate the changes of oxidative damage and total antioxidant capacity in the early development of Carassius auratus gibelio and the effects of adding vitamin C (Vc) on them. The contents of malondialdehyde (MDA), protein carbonyl (PCA), activity of lactate dehydrogenase (LDH) and total antioxidant capacity (T-AOC) in the early development of P. giganteum were determined by biochemical methods. (1) The hatching rate and survival rate of larvae in Vc solution at 30 mg / L were the highest, which were 16.2% and 17.8% higher than those in control group (Vc concentration 0 mg / L) respectively; (2) (3) 30 mg / L of Vc group (P <0.05), the content of malondialdehyde (MDA), the content of carbonyl group and the activity of lactate dehydrogenase all showed the tendency of “increasing - decreasing” (P <0.05), and the total antioxidant capacity was significantly higher than that of the control group (P <0.05). The contents of malondialdehyde, protein carbonyl and lactate dehydrogenase were significantly lower than those of the control group. The results showed that: (1) The oxidative damage was more serious in the embryos of P. glabra than in the embryo; (2) The proper concentration of vitamin C could enhance the antioxidant capacity of embryos and larvae, reduce the oxidative damage to embryos and larvae, Its hatching rate and survival rate.
