目的探讨高同型半胱氮酸血症(Hhcy)勃起功能下降大鼠阴茎海绵体内硫化氢(H_2S)信号通路及其作用。方法将20只2月龄雄性健康wistar大鼠适应性喂养1周后随机分为Hhcy组(H组)与对照组(C组),每组10只。给予H组(生理盐水3mL/次+蛋氨酸1g·kg~(-1)·d~(-1))和C组(生理盐水3mL/次)早晚各1次灌胃并持续6周。6周后采血检测大鼠的血清睾酮、血浆同型半胱氨酸(Hcy)和H_2S的浓度。阴茎海绵体内压/平均颈动脉压(ICP_(max)/MAP)测定后行阴茎海绵体H_2S含量及生成率检测,用Western blot和免疫组化法分析阴茎海绵体中胱硫醚-β-合成酶(CBS)与胱硫醚-γ-裂解酶(CSE)的含量与表达。结果大鼠血清睾酮值C组(463.56±142.19)nmol/L,H组(352.76±120.01)nmol/L,两组比较差异无统计学意义(P>0.05)。H组血浆Hcy浓度较C组明显升高[(27.34±2.49)μmol/Lys(11.41±1.50)μmol/L],差异具统计学意义(P<0.05),而其ICPmax/MAP(0、3、5V)均显著低于C组,差异具统计学意义(P<0.05)。血浆H_2S浓度、阴茎海绵体内H_2S的含量与生成率比较,H组[(13.36±1.75)μmol/L、(57.54±1.99)nmol/mg prot、(1.38±0.24)nmol/(mg·min)]较C组[(22.44±1.53)μmol/L、(88.63±2.05)nmol/mg prot、(4.53±0.33)nmol/(mg·min)]均显著降低,差异均具统计学意义(P<0.05)。阴茎组织中H组CSE和CBS的蛋白含量较C组明显减少,差异均具统计学意义(P<0.05)。结论阴茎海绵体内CSE和CBS含量减少和表达减弱,内源性H_2S含量降低,这可能是Hhcy大鼠阴茎勃起功能下降的原因之一。
Objective To investigate the effect of Hhcy erectile function on hydrogen sulfide (H2S) signal pathway in the penis of rats with hyperhomocysteinemia. Methods Twenty healthy male Wistar rats, aged 2 months, were fed for 1 week and then randomly divided into Hhcy group (H group) and control group (C group), with 10 rats in each group. The rats in group H (3mL / time + methionine 1g · kg -1 d -1) and group C (3mL / time saline) were given intragastrically once daily for 6 weeks. Six weeks after the blood test, serum testosterone, plasma homocysteine (Hcy) and H 2 S concentrations were measured. The contents and formation rate of H 2 S in the penis were measured after intracapsular pressure / mean carotid pressure (ICP_ (max) / MAP), and the cystathionine-β-synuclein in the penis was analyzed by Western blot and immunohistochemistry CBS and cystathionine-γ-lyase (CSE) content and expression. Results The level of serum testosterone in group C was (463.56 ± 142.19) nmol / L and that in group H was (352.76 ± 120.01) nmol / L, there was no significant difference between the two groups (P> 0.05). The plasma concentration of Hcy in group H was significantly higher than that in group C [(27.34 ± 2.49) μmol / Lys (11.41 ± 1.50) μmol / L], the difference was statistically significant (P <0.05) , 5V) were significantly lower than C group, the difference was statistically significant (P <0.05). Conclusions The reduction and expression of CSE and CBS in the penis and the decrease of endogenous H 2 S content may be one of the reasons for the decrease of erectile function in Hcy rats.
