目的 :探讨西比灵对高血压大鼠急性脑缺血再灌注后 HSP70 表达的影响及其脑保护作用。方法 :将大鼠用双肾动脉狭窄术制成高血压大鼠模型 ,喂养 2个月。随机分为两组 :测体积组 :再分为西比灵干预组、缺血再灌注组和假手术组。HSP70组 :也分为西比灵干预组、缺血再灌注组和假手术组。除假手术组大鼠外 ,其它大鼠均制成大脑中动脉闭塞 (MCAO)模型 ,MCAO时间均为 2 h。再灌注时点 :测体积组为 6 h;HSP70 组为 1 d。测体积组将鼠脑经 TTC染色后 ,用计算机病理图象分析仪测量出梗死面积 ,根据梯形法则计算出梗死体积及梗死体积比。测 HSP70 组 ,经免疫组化染色后用病理图象分析仪测出 HSP70阳性细胞数。结果 :(1 )西比灵组 HSP70 的表达高于缺血再灌注组 (P<0 .0 5 ) ;(2 )西比灵组脑梗死体积比明显小于缺血再灌注组 (P<0 .0 5 )。结论 :西比灵能促进 HSP70 的表达 ,使梗死体积减小 ,对缺血再灌注引起的受损脑组织有保护作用
Objective: To investigate the effect of xibeiling on HSP70 expression and its neuroprotective effect after acute cerebral ischemia-reperfusion in hypertensive rats. Methods: The rats were made hypertensive rat model by renal artery stenosis and fed for 2 months. Randomly divided into two groups: test volume group: sub-divided into the West Bailing intervention group, ischemia reperfusion group and sham operation group. HSP70 group: also divided into the West Billy intervention group, ischemia reperfusion group and sham operation group. All the rats except the sham operation group were given middle cerebral artery occlusion (MCAO) model, and the MCAO time was 2 h. At the time of reperfusion, the volume of test group was 6 h and the HSP70 group was 1 d. The volume of rats in the test group was stained with TTC, and the area of infarction was measured by computerized pathological image analyzer. The volume of infarction and infarction volume were calculated according to the trapezoidal rule. HSP70 group was measured, the number of HSP70 positive cells was measured by immunohistochemical staining with pathological image analyzer. Results: (1) The expression of HSP70 in Sibelium group was significantly higher than that in ischemia-reperfusion group (P <0.05); (2) The volume fraction of cerebral infarction in Xibiling group was significantly lower than that in ischemia-reperfusion group .0 5). Conclusion: Sibelium can promote the expression of HSP70, reduce infarct size and protect brain tissue damaged by ischemia-reperfusion
