目的 :探讨复方苦参水溶液体外对结肠癌细胞的影响。方法 :应用MTT法 ,生长曲线 ,荧光染色 ,流式细胞仪 ,透射电镜技术研究复方苦参水溶液体外对SW 480细胞的增殖抑制及诱导凋亡作用。结果 :复方苦参水溶液体外对SW 480细胞有明显的抑制作用 ,而且这种抑制作用呈现浓度和时间的依赖性。 30mg/ml中药作用SW 480细胞 48小时后 ,光镜及电镜下可见胞核固缩 ,荧光染色增强 ,胞核碎裂 ,凋亡小体形成等凋亡形态学变化。流式细胞仪检测到亚G1峰 ,其凋亡率为 11 13%。细胞周期分析发现 ,经 30mg/ml药物处理 48小时后 ,S期细胞由对照组的 2 5 41%上升到 41 2 7% (P <0 0 1) ,G2 /M期细胞由 6 74%降至 1 35 % (P <0 0 1)。结论 :复方苦参水溶液体外能够抑制SW 480细胞增殖并且在一定浓度范围内诱导其凋亡 ,诱导凋亡可能是其发挥抗肿瘤作用的重要机制之一。
Objective: To explore the effect of compound Kushen Aqueous Solution on colon cancer cells in vitro. METHODS: MTT assay, growth curve, fluorescence staining, flow cytometry, and transmission electron microscopy were used to study the effects of compound Kushen Aqueous Solution on proliferation inhibition and apoptosis induction of SW 480 cells in vitro. RESULTS : The aqueous solution of Compound Radix Sophorae Radix was significantly inhibited in SW 480 cells in vitro and the inhibitory effect appeared concentration and time dependent. After treated with 30mg/ml Chinese herbal medicine SW 480 cells for 48 hours, pyknosis, fluorescent staining, nuclear fragmentation, apoptotic body formation and other apoptotic morphological changes were observed under light microscope and electron microscope. The sub-G1 peak was detected by flow cytometry and the apoptotic rate was 11 13%. Cell cycle analysis showed that after 48 hours of treatment with 30 mg/ml drug, S phase cells increased from 251 41% in the control group to 41 2 7% (P <0 01), and the G2 /M phase cells decreased by 6 74%. Conclusion : Compound Sophora flavescens aqueous solution can inhibit the proliferation of SW 480 cells in vitro and induce its apoptosis within a certain range of concentration. Induction of apoptosis may be one of the important mechanisms of its anti-tumor effect.
