目的通过观察左归丸含药血清对成骨细胞骨保护素(OPG)、细胞核因子-κB受体活化因子配基(RANKL)mRNA表达的影响,探讨其治疗骨质疏松症的作用机制。方法体外分离、培养成骨细胞,实验分为3组:正常血清组、卵巢切除血清组、卵巢切除含药血清组。采用原位杂交法,检测成骨细胞OPG、RANKL mRNA的表达。结果卵巢切除血清组与正常血清组相比,成骨细胞OPG mRNA的表达显著下调,而RANKL mRNA表达明显上调;卵巢切除含药血清组与卵巢切除血清组相比,成骨细胞OPG mRNA的表达明显上调,RANKL mRNA的表达明显下调;而与正常血清组相比,无显著性差异。结论在去势状态下,左归丸含药血清一方面直接抑制RANKL的分泌,使破骨细胞活性降低;另一方面促进成骨细胞分泌OPG,使之与RANKL的结合增多,进而使破骨细胞活性降低,从而达到治疗骨质疏松的目的。
Objective To observe the effects of Zuogui Pill-containing serum on osteoprotegerin osteoprotegerin (OPG) and nuclear factor-κB receptor ligand (RANKL) mRNA expression, and to explore the mechanism of osteoporosis treatment. Methods Osteoblasts were isolated and cultured in vitro. The experiments were divided into 3 groups: normal serum group, oophorectomized serum group, and ovariectomy-containing serum group. The expression of OPG and RANKL mRNA in osteoblasts was detected by in situ hybridization. Results OPG mRNA expression in osteoblasts was significantly down-regulated and RANKL mRNA expression was significantly up-regulated in ovariectomized serogroups compared with normal sera. OPG mRNA expression in osteoblasts was significantly higher in ovariectomized sera than in ovariectomized serogroups. Obviously up-regulated, RANKL mRNA expression was significantly down-regulated; compared with normal serum group, there was no significant difference. Conclusion In the state of castration, Zuogui Pill-containing serum directly inhibits the secretion of RANKL on one hand, and reduces the activity of osteoclasts. On the other hand, it promotes osteoblasts to secrete OPG, which increases the binding of RANKL to osteoclasts. Cell activity is reduced to achieve osteoporosis treatment.
