【目的】探讨对核桃基腐病(Pythium oligandrum Drechsler)有较好抑制效果的菌株Gy-11的最优发酵条件,提高其次生代谢产物的产量。【方法】开展了最适培养基和最优碳氮源的筛选,以菌体生物量和发酵液对病原菌的抑菌活性为指标,采用单因素方法对菌株的发酵条件进行优化。【结果】菌株Gy-11在LB培养基和NB培养基中生长量最大;生长所需的最佳碳源为葡萄糖,最佳氮源为蛋白胨;菌株Gy-11最佳发酵条件为温度25℃,p H 7.0,装液量50 m L/250m L,接种量体积分数为6%,转速为180 r·min~(-1)。【结论】明确了菌株Gy-11的最适碳氮源和最优发酵条件为进一步研发生防制剂提供理论基础。
【Objective】 The purpose of this study was to investigate the optimum fermentation conditions of Gy-11 strain Pythium oligandrum Drechsler and to increase its yield of secondary metabolites. 【Method】 The optimum culture medium and optimal carbon and nitrogen sources were screened. The bacterial biomass and the bacteriostasis of the fermentation broth to the pathogenic bacteria were used as indexes. The fermentation conditions of the strains were optimized by single factor method. 【Result】 The strain Gy-11 had the highest growth in LB medium and NB medium. The best carbon source for growth was glucose and the best nitrogen source was peptone. The optimal fermentation conditions of strain Gy-11 were 25 ℃ , p H 7.0, liquid volume 50 m L / 250m L, inoculum volume fraction 6%, speed 180 r · min -1. 【Conclusion】 The optimum carbon source and optimal fermentation conditions of strain Gy-11 were clarified to provide the theoretical basis for further research on biocontrol agents.
