目的:建立腰椎间盘退变的大鼠模型,观察淫羊藿苷对其Ⅱ型胶原蛋白和基质金属蛋白酶表达的影响,从而判断淫羊藿苷是否能改善椎间盘退变。方法:将60只Wistar大鼠随机分为3组:假手术组、对照组、淫羊藿苷组,每组20只。其中对照组、淫羊藿苷组采用纤维环穿刺法建立大鼠椎间盘退变模型,假手术组只做切开,不做穿刺。建立模型后1周,淫羊藿苷组以6.0 g/(kg·d),空白对照组和假手术组予以等量的生理盐水灌胃,持续8周。结果:RT-PCR的方法定量分析各组的Ⅱ型胶原蛋白m RNA和基质金属蛋白酶m RNA的表达。对照组的Ⅱ型胶原蛋白m RNA的表达明显低于假手术组,基质金属蛋白酶m RNA的表达明显高于假手术组。淫羊藿苷组的Ⅱ型胶原蛋白m RNA的表达明显高于对照组,基质金属蛋白酶m RNA的表达明显低于对照组。结论:淫羊藿苷能明显改善椎间盘的退变。
OBJECTIVE: To establish a rat model of lumbar disc degeneration and observe the effect of icariin on the expression of collagen type Ⅱ and matrix metalloproteinases (MMPs), so as to determine if icariin can improve the degeneration of intervertebral discs. Methods: Sixty Wistar rats were randomly divided into three groups: sham operation group, control group and icariin group, 20 rats in each group. The control group and icariin group were treated by fibrocystic puncture to establish the model of intervertebral disc degeneration in rats. Sham-operated group only incision and did not puncture. One week after model establishment, icariin group was administrated with normal saline (6.0 g / (kg · d)) and blank control group and sham operation group for 8 weeks. Results: RT-PCR was used to quantitatively analyze the expression of type II collagen mRNA and matrix metalloproteinase mRNA in each group. The expression of type Ⅱ collagen m RNA in the control group was significantly lower than that in the sham-operation group, and the expression of MMP-2 mRNA in the control group was significantly higher than that in the sham-operated group. Icariin group of collagen type m RNA expression was significantly higher than the control group, the expression of matrix metalloproteinase m RNA was significantly lower than the control group. Conclusion: Icariin can significantly improve the degeneration of intervertebral disc.
