目的建立菏泽地区丹凤牡丹花瓣的HPLC指纹图谱,并将其应用于不同品种牡丹花的质量评价,同时测定不同品种牡丹花中8种成分的量,为科学评价牡丹花质量提供依据。方法采用纳微Nano-Micro C_(18)(250 mm×4.6 mm,5μm)色谱柱;以乙腈-0.1%甲酸水溶液为流动相,梯度洗脱,体积流量1.0 mL/min,检测波长270 nm,柱温25℃。运用中药色谱指纹图谱相似度评价系统进行分析。结果 6批次丹凤牡丹花瓣样品得到的指纹图谱相似度大于0.993,标定共有峰14个,并对其中8个色谱峰进行了归属。同时对6批不同品种牡丹样品与丹凤牡丹进行相似度分析比较,整体相似度较高。同时对牡丹花瓣中没食子酸、没食子酸甲酯、山柰酚-3-O-β-D-吡喃葡萄糖苷(紫云英苷)、芹菜素-7-O-β-D-吡喃葡萄糖苷(大波斯菊苷)、野漆树苷、二氢山柰酚-7-O-β-D-吡喃葡萄糖苷、芍药苷、氧化芍药苷8种主要代谢产物进行了定量测定,其质量分数分别在0.05~1.41、1.01~4.41、0.70~5.55、4.60~18.30、10.05~33.87、0.09~1.76、3.16~11.12、0.85~2.02 mg/g。结论 HPLC指纹图谱结合定量测定能全面反映内在质量,该方法简便可行、快速准确,为牡丹花质量评价提供参考。
OBJECTIVE To establish the HPLC fingerprint of the petals of Danfeng peony in Heze area and to apply it to the quality evaluation of different varieties of peony flowers and to determine the content of eight components in different varieties of peony flower and provide scientific basis for the scientific evaluation of the quality of peony flowers. Methods The mobile phase consisted of acetonitrile-0.1% formic acid aqueous solution with gradient elution at a flow rate of 1.0 mL / min. The detection wavelength was set at 270 nm. Column temperature 25 ℃. The similarity analysis system of chromatographic fingerprints of traditional Chinese medicine was used for analysis. Results The fingerprints similarity of the samples from 6 batches of Danfeng peony was greater than 0.993, the total number of peaks was 14 and the eight peaks were assigned. At the same time, six groups of different varieties of peony samples and Danfeng Peony similarity analysis and comparison, the overall similarity is high. In the meantime, the effects of gallic acid, methyl gallate, kaempferol-3-O-β-D-glucopyranoside (Astragalin), apigenin-7-O-β-D-glucopyranose Glycosides (Cosmos glycosides), wild laccoside, dihydro-kaempferol -7-O-β-D-glucopyranoside, paeoniflorin, paeoniflorin eight kinds of major metabolites were determined, the mass fraction Respectively in the range of 0.05-1.41, 1.01-4.41, 0.70-5.55, 4.6O- 18.30, 10.05-33.87, 0.09-1.76, 3.16-11.12 and 0.85-2.02 mg / g. Conclusion HPLC fingerprinting combined with quantitative determination can fully reflect the intrinsic quality. The method is simple and feasible, rapid and accurate, and provides a reference for the quality evaluation of peony flowers.
