[方法]16HBE细胞经0.0625~1.0 mg/L剂量的克菌丹染毒,通过观察染毒细胞形态及恶性表型鉴定,检测克菌丹诱导16HBE细胞的转化。[结果]克菌丹转化第30代细胞生长速度增快,对conA凝集敏感性增强,失去贴壁生长依赖性,电镜下可见明显的形态学改变,裸鼠体内形成肿块为鳞状上皮细胞癌。[结论]克菌丹可直接诱导16HBE细胞发生恶性转化,该模型的建立为克菌丹对健康损伤效应研究及其评价奠定了基础。
[Method] 16HBE cells were treated with captan at a dose of 0.0625 ~ 1.0 mg / L. The morphological changes and malignant phenotypes of infected cells were observed to determine the conversion of 16HBE cells. [Result] The growth of captan at the 30th generation was accelerated, the sensitivity to conA agglutination was enhanced, and the dependence on adherent growth was lost. The obvious morphological changes were observed under electron microscope. The nude mice formed squamous cell carcinoma . [Conclusion] Captan can directly induce the malignant transformation of 16HBE cells. The establishment of this model lays the foundation for the study and evaluation of the effect of captan on health damage.
