目的:针对不同生长时期人参中水溶性蛋白的含量进行比较。方法:以5年生人参为供试材料,应用Bradford蛋白质定量试剂盒测定水溶性蛋白含量,采用聚丙烯酰胺凝胶电泳(SDS-PAGE)法得到人参水溶性蛋白的电泳胶片,并通过凝胶成像分析软件和Origin 7.5软件生成指纹图谱,导出重要技术参数,分析比较不同生长时期人参中水溶性蛋白的分布及含量差异。结果:不同生长时期的人参样品均显示12条水溶性蛋白谱带,且在人参果后参根生长中期直至枯萎期,水溶性蛋白含量达到最高。结论:人参水溶性蛋白的含量在不同生长时期存在较大差异,与人参生长的形态建成密切相关。
Objective: To compare the contents of water-soluble proteins in ginseng at different growth stages. Methods: The 5-year-old ginseng was used as test material, and the content of water-soluble protein was determined by Bradford protein quantitative kit. The electrophoresis film of ginseng water-soluble protein was obtained by polyacrylamide gel electrophoresis (SDS-PAGE) Analysis software and Origin 7.5 software generate fingerprints and derive important technical parameters to analyze and compare the differences in the distribution and content of water-soluble proteins in different growth stages. RESULTS: Twelve water-soluble protein bands were observed in different growth stages of ginseng samples, and the highest content of water-soluble protein was observed in the middle of ginseng root growth period until wilt. CONCLUSION: The content of water-soluble protein in ginseng is quite different at different growth stages, which is closely related to the morphogenesis of ginseng growth.
