目的:研究脂多糖诱导兔股动脉外膜炎症对内膜增生的影响,探讨外膜炎症对内膜增生影响的分子机制。方法:成年新西兰大耳白兔30只,术前均喂以高脂饮食2周。两侧的股动脉随机分为脂多糖组和对照组。于术后1h、3d和2周分别处死10只动物获得股动脉标本,观察形态学变化和血管内膜增生情况,并用免疫组化方法观察P-选择素在动脉壁的表达。结果:术后3d和2周时,脂多糖组血管外膜和内膜面有明显的炎症浸润。术后2周脂多糖组内膜面积(0.95±0.10)mm2和对照组(0.73±0.07)mm2相比,差异有统计学意义(P<0.05)。2组P-选择素在术后1h在动脉壁均无表达;在术后3d时脂多糖组血管外膜细胞和内皮细胞在胞浆中出现P-选择素阳性表达,而在对照组无明显阳性表达。结论:脂多糖诱导的血管外膜炎症可导致兔股动脉内膜增生,P-选择素参与了这一过程。
Objective: To investigate the effect of lipopolysaccharide-induced inflammation of rabbit’s femoral artery on intimal hyperplasia and to explore the molecular mechanism of the effect of adventitial inflammation on intimal hyperplasia. Methods: Thirty New Zealand white rabbits were infused with high-fat diet for 2 weeks before operation. Both sides of the femoral artery were randomly divided into lipopolysaccharide group and control group. Ten animals were sacrificed at 1, 3 and 2 weeks after surgery to obtain the femoral artery. Morphological changes and intimal hyperplasia were observed. The expression of P-selectin in the artery wall was observed by immunohistochemistry. Results: At 3 and 2 weeks after operation, there was obvious inflammatory infiltration in the adventitia and intima of the lipopolysaccharide group. The area of intima of lipopolysaccharide group (0.95 ± 0.10) mm2 at 2 weeks after operation was significantly higher than that of control group (0.73 ± 0.07) mm2 (P <0.05). P-selectin was not expressed in the arterial wall at 1h after operation in both groups. P-selectin expression was observed in the cytoplasm of LPS group and 3d in the LPS group, but not in the control group Positive expression. CONCLUSIONS: LPS-induced inflammation of the adventitia leads to intimal hyperplasia of rabbit femoral artery, and P-selectin is involved in this process.
