为了解精子 DNA 损伤在男性生殖健康中的意义,对421名男性的精子用计算机辅助分析系统常规检测后做单细胞凝胶电泳试验(SCGE),根据精子细胞核 DNA 损伤程度分为5级。结果表明,精子密度的不同,彗星样细胞发生率有明显的改变,当密度<20×10~6/ml 时,彗星样细胞发生率特别是Ⅱ级、Ⅲ级显著升高;在不动的 d 级精子中,Ⅰ级彗星样细胞发生率占5.39%,Ⅱ、Ⅲ级彗星样细胞发生率显著升高,与 a 级精子相比差异有显著意义(P<0.05)。结论:SCGE 试验检测精子 DNA 链断裂可评价精子的质量和损伤,是男性生殖健康评估的又一较为可靠的检测手段。
To understand the significance of sperm DNA damage in male reproductive health, 421 male sperm were routinely tested with a computer-assisted analysis system and then performed a single cell gel electrophoresis assay (SCGE), which was classified into 5 grades based on the degree of sperm DNA damage. The results showed that the incidence of comet-like cells was significantly changed with different sperm densities. When the density was less than 20×10 6 /ml, the incidence of comet-like cells was particularly high in grades II and III. Among d-type spermatozoa, the incidence of I-type comet-like cells accounted for 5.39%, and the incidence of II-III-like comet-like cells increased significantly, which was significantly different from that of a-type spermatozoa (P<0.05). Conclusion: The SCGE test to detect sperm DNA strand breaks can be used to evaluate the quality and damage of sperm. It is another more reliable detection method for male reproductive health assessment.
