禁欲期正常志愿者精液中生精细胞经直接低渗处理,制备有丝分裂和减数分裂染色体的新方法及形态学观察结果为:1.正常人精液中脱落的精原细胞经直接低渗处理后,有丝分裂染色体主要为粗短形中期染色体(二倍体,染色均匀)。来源于初级/次级精母细胞减数分裂的染色体多为细长形(单倍体、二倍体均有,因期而异)。2.本法可获得较多分裂相,分裂指数平均为11.5%。二倍体与单倍体比例约为1:2(222/446),三倍体(0.58%,4/689)和四倍体(2.5%,17/689)也可看到。3.有较多的第一次减数分裂前期分裂相。包括有联会复合体的偶线期、四分体的粗线期、出现姐妹染色单体分离和交叉的双线期以及染色体明显缩短的终变期。后期Ⅰ、末期Ⅰ和第二次减数分裂的图像较少见。若排精间隔少于24h,生精细胞一般不易制备分裂期染色体;间隔24h后,随间隔时间延长染色体分裂相也增多。4.第一次减数分裂前期生精细胞性染色体可见长臂端部有"尾巴样细丝结构",此期以后的细胞未见此结构。
Abstinence normal volunteer sperm germ cells by direct hypotonic treatment of mitosis and meiosis chromosome preparation of new methods and morphological observations as follows: 1. Normal semen sperm cells shed by the direct hypotonic treatment, the mitotic chromosomes are mainly coarse and short metaphase chromosomes (diploid, uniform staining). Chromosomes derived from meiosis of primary / secondary spermatocytes are mostly elongated (haploid, diploid, period-specific). 2. This method can get more split phase, split index averaged 11.5%. The diploid to haploid ratios were also seen at 1: 2 (222/446), triploid (0.58%, 4/689) and tetraploid (2.5%, 17/689) . 3. There are more first premenstrual split phase. Including the association of complex even line period, quarter of the thick line, sister chromatid separation and cross the two-line phase and the chromosome significantly shortened the terminal stage. Late I, late I and second meiosis images are rare. If the row of semen less than 24h, spermatogenic cells generally not easy to prepare mitotic chromosomes; after a lag interval of 24 hours, with the extension of interphase chromosome cleavage phase also increased. 4. The first meiotic spermatogenic chromosomes showed a “tail-like filament structure” at the end of the long arm, and the cells did not show this structure after this period.
