目的观察n-3多不饱和脂肪酸(n-3PUFAs)对大鼠大肠癌及Caspase-3蛋白表达的影响。方法将60只SD大鼠随机分成对照组和实验组,实验组给予n-3PUFAs灌胃,对照组给予等量的生理盐水灌胃,两组均使用N-甲基-N-亚硝基脲(MNU)进行灌肠。16周后解剖大鼠,观察肿瘤情况,HE切片明确肿瘤的病理,气相色谱测定红细胞n-3PUFAs的浓度,免疫组化检测癌组织及正常组织Caspase-3蛋白的表达。结果实验组大鼠的红细胞n-3PUFAs浓度高于对照组(P<0.01),肿瘤发生率低于对照组(P<0.05),Caspase-3蛋白表达高于对照组(P<0.05),差异均有统计学意义。结论 n-3PUFAs能够减少MNU诱导大鼠大肠癌的发生率,可能机制是增加癌细胞凋亡。
Objective To observe the effect of n-3 polyunsaturated fatty acids (n-3 PUFAs) on the expression of Caspase-3 and protein in colorectal cancer in rats. Methods Sixty Sprague-Dawley rats were randomly divided into control group and experimental group. The experimental group was given n-3PUFAs intragastrically and the control group was given normal saline intragastrically. N-methyl-N-nitrosourea (MNU) for enema. Twenty-six weeks later, the rats were dissected and the tumor was observed. The pathology of tumor was determined by HE section. The concentration of erythrocyte n-3PUFAs was determined by gas chromatography. The expression of Caspase-3 protein in cancer tissue and normal tissue was detected by immunohistochemistry. Results The concentration of erythrocyte n-3 PUFAs in the experimental group was significantly higher than that in the control group (P <0.01), the incidence of tumor was lower than that of the control group (P <0.05), and the expression of Caspase-3 protein was higher than that of the control group All have statistical significance. Conclusions n-3PUFAs can reduce the incidence of colorectal cancer induced by MNU in rats. The possible mechanism is to increase the apoptosis of cancer cells.
