本研究将齐墩果酸(OA)与5-氟尿嘧啶(5-Fu)联合作用于A549细胞,用HPLC法测定有无齐墩果酸作用后细胞内外5-Fu的浓度,观察药物浓度的变化情况。结果显示当未给予OA时,刚开始5-Fu浓度与作用时间呈正相关,但增加到一定浓度的时候,其细胞内的浓度基本没有显著的变化,由此可能出现耐药性的情况。当给予OA后,细胞内的药物浓度一开始时就出现比较高的浓度,而随着作用时间的增长,细胞内的浓度并没有增加,呈现一个相对比较稳定的范围,由此推测可能是OA影响了细胞膜对5-Fu的转运。由此推测齐墩果酸可能会延长5-氟尿嘧啶的耐药性产生时间,并能与5-氟尿嘧啶具有协同的的抗癌细胞效果。
