目的:确定抗人CD38分子单克隆抗体(mAb)1C6识别的抗原表位所在区域。方法:分别构建缺失人CD38不同表位的重组质粒,用IPTG诱导表达融合蛋白。对表达产物进行SDS-PAGE蛋白电泳及Westernblot分析。结果:1C6不能识别缺失第220~241位氨基酸的D4片段,而对其他的缺失突变体均可识别。结论:mAb1C6抗体识别的抗原表位位于CD38分子胞外段第220~241位氨基酸。
OBJECTIVE: To determine the region of antigenic epitope recognized by anti-human CD38 monoclonal antibody (mAb) 1C6. Methods: Recombinant plasmids with different epitopes of human CD38 were constructed respectively, and induced by IPTG to express the fusion protein. The expressed product was subjected to SDS-PAGE protein electrophoresis and Western blot analysis. Results: 1C6 did not recognize the D4 fragment deleted from amino acids 220-241, but all other deletion mutants were recognized. Conclusion: The epitope recognized by mAb1C6 antibody is located at amino acids 220-241 of extracellular domain of CD38.
