诱导性多能干细胞(iPS)是以反转录病毒为载体将胚胎特异性转录因子导入体细胞,使其诱导分化为具有胚胎干细胞(ESCs)特性的细胞。这种多能干细胞在细胞形态、增殖速率、致瘤性、基因表达以及形成嵌合小鼠的能力上与ESCs有许多相似之处,将来可能成为ESCs在临床应用中的替代。现对iPS细胞相关的几种转录因子及其在重编程过程中的作用,以及转导基因的选择予以综述。
Induced pluripotent stem cells (iPS) are retroviral vectors for the introduction of embryo-specific transcription factors into somatic cells, which are induced to differentiate into embryonic stem cells (ESCs). This pluripotent stem cell has many similarities with ESCs in terms of cell morphology, proliferation rate, tumorigenicity, gene expression, and the ability to form chimeric mice and may in the future be an alternative to ESCs for clinical use. Several transcription factors related to iPS cells and their roles in reprogramming, as well as the selection of transduction genes, are reviewed.
