目的:探讨4Gy 137Csγ射线一次性全身照射小鼠免疫细胞对LPS刺激反应性的远期影响。方法:将C57BL/6实验小鼠分为假照射组和照射组,照射组给予4 Gy照射。照射10周后小鼠按组别分别提前24 h和1 h腹腔注射LPS(20mg/kg),对照组注射生理盐水。取外周血进行白细胞计数及CD4、CD8、B220细胞比例检测,脾脏与胸腺称重,计算脏器指数。冲洗单侧股骨进行有核细胞计数。结果:与假照射对照组小鼠比较,照射对照组小鼠的CD4,CD8细胞比例和脾脏、胸腺指数显著升高,B细胞比例显著下降,差异有统计学意义(P<0.05)。LPS刺激1 h后,照射组小鼠的CD4细胞较假照射组小鼠显著降低,差异有统计学意义(P<0.05)。LPS24 h后,与假照射组小鼠相比,照射组小鼠CD8细胞比例显著降低,胸腺指数显著升高,差异有统计学意义(P<0.05)。结论:小鼠4 Gy照射10周后,免疫系统尚未完全恢复;在接受LPS刺激后,两组小鼠反应也有差异。辐射对小鼠免疫系统损伤的远期影响有待进一步研究。
Objective: To investigate the long-term effects of 4Gy 137Cs γ-ray irradiation on immune responses to LPS stimulation in mice. Methods: C57BL / 6 mice were divided into sham irradiation group and irradiation group. The irradiation group was given 4 Gy irradiation. Mice were injected intraperitoneally with LPS (20 mg / kg) 24 h and 1 h respectively after 10 weeks of irradiation. The control group was injected with saline. Peripheral blood was taken for white blood cell count and CD4, CD8, B220 cells were detected, the spleen and thymus were weighed and the organ index was calculated. Routine unilateral femoral nucleated cell count. Results: Compared with the sham-irradiated control mice, the proportion of CD4 and CD8 cells, the spleen and thymus index and the percentage of B cells in irradiated control group were significantly decreased (P <0.05). After 1 h of LPS stimulation, the CD4 cell level in irradiated mice was significantly lower than that in sham-irradiated mice (P <0.05). Compared with sham-irradiated mice, the proportion of CD8 cells in irradiated mice was significantly decreased and the thymus index was significantly increased at 24 h after LPS injection (P <0.05). CONCLUSION: After 4 weeks of irradiation, the immune system has not yet recovered completely. After LPS stimulation, the responses of the two groups of mice are also different. The long-term effects of radiation on the immune system damage in mice are for further study.
