目的探讨艾迪注射液中刺五加提取物对斑蝥素的增效、减毒作用。方法采用MTT法考察刺五加提取物对斑蝥素引起的肿瘤细胞(SMMC-7721肝癌细胞)和正常大鼠肾NRK细胞的细胞毒作用;采用流式细胞术检测SMMC-7721和NRK细胞凋亡情况。用斑蝥素制作大鼠急性肾损伤模型,考察刺五加提取物对肾损伤大鼠的保护作用。结果刺五加提取物可增强斑蝥素对SMMC-7721肝癌细胞增殖的抑制作用,降低斑蝥素对NRK细胞的细胞毒作用(P<0.05或<0.01);刺五加提取物能增强斑蝥素引起SMMC-7721细胞凋亡,抑制NRK细胞凋亡(P<0.05或<0.01)。刺五加提取物可降低斑蝥素所致肾损伤大鼠的尿素氮、肌酐含量,减少尿液中尿蛋白含量(P<0.05或<0.01)。结论艾迪注射液中刺五加提取物对斑蝥素有增效、减毒作用。
Objective To investigate the synergistic and attenuated effects of acanthopanax senticosus extract in Aidi Injection on cantharidin. Methods MTT assay was used to investigate the cytotoxic effect of Acanthopanax senticosus extract on tumor cells (SMMC-7721 hepatoma cells) induced by cantharidin and normal rat renal NRK cells. Flow cytometry was used to detect the apoptosis of SMMC-7721 and NRK cells Happening. Acute kidney injury model was induced by cantharidin in rats, and the protective effect of Acanthopanax senticosus extract on renal injury in rats was investigated. Results Acanthopanax senticosus extract could enhance the inhibitory effect of cantharidin on the proliferation of SMMC-7721 hepatocarcinoma cells and reduce the cytotoxic effect of cantharidin on NRK cells (P <0.05 or <0.01). Acanthopanax senticosus extract can enhance the induction of cantharidin SMMC-7721 cells could inhibit apoptosis of NRK cells (P <0.05 or <0.01). Acanthopanax extract can reduce the cantharidin-induced renal injury rats urea nitrogen, creatinine, reduce urinary proteinuria (P <0.05 or <0.01). Conclusion Aidi injection acanthopanax senticosus extract has a synergistic and attenuated effect.
