目的探讨蛇毒精氨酸酯酶Agkihpin对人鼻咽癌CNE-2细胞系中表皮钙粘素(E-CD)表达的影响,并试图阐明Agkihpin抑制人鼻咽癌CNE-2细胞的机制。方法用不同浓度的Agkihpin处理鼻咽癌细胞株72 h后,应用免疫细胞化学、RT-PCR法检测E-CD在CNE-2细胞中的表达。结果不同浓度Agkihpin作用CNE-2细胞72 h后E-CD表达均降低,并呈现出一定的浓度依赖效应,显示Agkihpin可显著下调CNE-2细胞中E-CD表达。各加药组与不加Agkihpin组比较,差异具有统计学意义(P<0.05)。结论在人低分化鼻咽癌CNE-2细胞系中,Agkihpin能抑制E-CD的表达,并且随浓度的增大有明显的抑制作用。
Objective To investigate the effect of Agkihpin on the expression of epidermal cadherin (E-CD) in human nasopharyngeal carcinoma cell line CNE-2 and to elucidate the mechanism by which Agkihpin inhibits human nasopharyngeal carcinoma CNE-2 cells. Methods Nasopharyngeal carcinoma cell lines were treated with different concentrations of Agkihpin for 72 h. Immunocytochemistry and RT-PCR were used to detect the expression of E-CD in CNE-2 cells. Results The E-CD expression of CNE-2 cells treated with different concentrations of Agkihpin for 72 h decreased and showed a concentration-dependent effect. The results showed that Agkihpin significantly down-regulated the expression of E-CD in CNE-2 cells. Each dosing group compared with without Agkihpin group, the difference was statistically significant (P <0.05). Conclusion Agkihpin can inhibit the expression of E-CD in human poorly differentiated nasopharyngeal carcinoma CNE-2 cell line, and significantly inhibit the growth of CNE-2 cells with increasing concentration.
