目的观察人胃腺癌裸鼠移植瘤125I粒子植入后近期放射损伤。方法建立人胃腺癌BGC-823细胞裸鼠皮下移植瘤模型,治疗组在肿瘤内将4粒表面活度为9.25MBq125I粒子平均植入到肿瘤中。对照组植入4粒无放射活性的粒子。观察60d,计算存活率、移植瘤抑瘤率的变化;荷瘤部位皮肤的放射损伤反应。应用光镜观察两组的病理组织学变化,并评价组织学病理反应。结果治疗期间实验组与对照组裸鼠存活率均为100%。实验组和对照组比较10d抑瘤率即差异显著(P<0.01);60d抑瘤率为96.8%。(30±5)d(4/10)裸鼠荷瘤部位皮肤出现不同程度的水肿及红斑,(55±3)d有(5/10)出现皮肤损伤,渐形成溃疡,溃疡直径为(7.46±3.24)mm,呈火山口状,基底部结痂,痂下可见脓性分泌物;其余肤色正常。光镜下可见溃疡基底部有瘤细胞碎片及炎性细胞浸润;病理学反应均为直肠癌消退分级标准1级。结论125I粒子组织间种植对人胃癌裸鼠皮下移植瘤有显著的治疗效果,同时皮肤的放射损伤发生率也较高。
Objective To observe the recent radiation injury after transplanted human gastric adenocarcinoma 125I seeds in nude mice. Methods Human gastric adenocarcinoma BGC-823 cells were transplanted subcutaneously in nude mice. The treatment group was implanted with an average of 4 particles with a surface activity of 9.25MBq125I in the tumor. The control group was implanted with 4 non-radioactive particles. After 60 days of observation, the survival rate and tumor growth inhibition rate were calculated. The radiation injury of the skin of tumor bearing part was observed. The pathological changes of the two groups were observed with light microscope, and the histopathological responses were evaluated. Results The survival rate of nude mice in experimental group and control group during treatment was 100%. The difference between the experimental group and the control group in 10 days was significant (P <0.01), and the inhibition rate was 96.8% at 60 days. (30 ± 5) d (4/10), there were different degree of edema and erythema on the skin of tumor-bearing part of nude mice. (5/10) skin lesions appeared on (55 ± 3) days and ulceration gradually formed. ± 3.24) mm, crater-like, basal scab, purulent secretions visible; the rest of the normal skin color. Light microscopic ulcer basaltic showed tumor cell debris and inflammatory cell infiltration; pathological reactions were remission grade 1 rectal cancer. Conclusion The implantation of 125I seeds has significant therapeutic effect on subcutaneous xenografts of human gastric cancer in nude mice, and the incidence of skin radiation injury is also high.
