本实验研究30%体表面积Ⅲ度烫伤大鼠的肝与骨骼肌细胞核体外转录活性,分别观察了伤后6、12、24、48和72小时转录活性的动态变化。 实验结果表明,烫伤后6小时大鼠肝细胞核[~3H]-UTP掺入达峰值,与对照组比较差异非常显著(P<0.001),24小时后迅速降低至对照组水平。而骨胳肌细胞核[~3H]-UTP掺入,在伤后12小时达峰值(P<0.001),是对照组水平的172%,并一直持续到72小时,转录活性仍在很高的水平上。 上述结果提示,创伤后肝与骨骼肌组织中蛋白质生物合成速度增加,可能与这些组织中细胞核转录活性升高有关。
In this study, the transcriptional activity of liver and skeletal muscle nuclei in 30% body surface area Ⅲ degree scalded rats were studied in vitro. The dynamic changes of transcriptional activity at 6, 12, 24, 48 and 72 hours after injury were observed. The experimental results showed that [~ 3H] -UTP incorporation reached the peak at 6 hours after scald in rats, which was significantly different from that in the control group (P <0.001), and rapidly decreased to the level of the control group after 24 hours. However, incorporation of [~ 3H] -UTP into skeletal muscle cells peaked at 12 hours post injury (P <0.001), which was 172% of the control level and lasted for 72 hours and the transcriptional activity was still very high on. The above results suggest that the increased protein biosynthesis in posttraumatic liver and skeletal muscle may be related to the increased nuclear transcriptional activity in these tissues.
