目的研究EGCG对体外培养子宫肌瘤细胞增殖和凋亡的作用,及对p-AKT和PTEN蛋白表达的影响。方法体外培养子宫肌瘤细胞,不同浓度EGCG干预后,应用四甲基偶氮唑蓝(MTT)法检测细胞增生活性;PI染色FCM法分析检测细胞周期变化及凋亡率;免疫细胞化学测定p-AKT和PTEN蛋白表达情况。统计学分析采用方差分析和t检验。结果 EGCG对体外培养子宫肌瘤细胞有增殖抑制作用(P<0.05),呈时间、浓度依赖性,以剂量依赖方式诱导子宫肌瘤细胞凋亡(P<0.05),上调PTEN表达,下调p-AKT表达。结论 EGCG通过上调PTEN表达、下调p-AKT表达抑制肌瘤细胞的体外增生。
Objective To study the effect of EGCG on the proliferation and apoptosis of uterine fibroids in vitro and its effect on the expression of p-AKT and PTEN. Methods Uterine fibroids were cultured in vitro. After different concentrations of EGCG intervention, the cell proliferation was detected by MTT assay. The cell cycle and apoptosis were detected by PI staining FCM. The expression of p -AKT and PTEN protein expression. Statistical analysis using analysis of variance and t test. Results EGCG could inhibit the proliferation of uterine fibroids in vitro (P <0.05) in a time-and dose-dependent manner, induce apoptosis of uterine leiomyoma cells in a dose-dependent manner (P <0.05), up-regulate the expression of PTEN and down- AKT expression. Conclusion EGCG can inhibit the proliferation of myoma cells in vitro by up-regulating the expression of PTEN and down-regulating the expression of p-AKT.
