在研究人类染色体的结构异常时,染色体标本制备的质量会直接影响着对染色体的分析。规范与量化操作步骤能保证染色体制备的重复性和一致性,确保得到数量较多、又易于分析的染色体。外周血染色体培养操作过程中影响因素多,如细胞培养基、秋水仙素、胰酶浓度、制片过程等。因此,本文对国内报道的染色体标本制备改良技术作一综述,以便得到高质量的中期染色体。
In studying the structural abnormalities of human chromosomes, the quality of chromosome preparation will directly affect the analysis of chromosomes. Normative and quantitative steps to ensure the reproducibility and consistency of chromosome preparation, to ensure that a larger number, but also easy to analyze chromosomes. There are many influencing factors in the process of culture of peripheral blood chromosomes, such as cell culture medium, colchicine, pancreatin concentration, film making process and so on. Therefore, this article reviews the reported improvement techniques of chromosome specimen preparation in order to obtain high-quality metaphase chromosomes.
