以尾叶桉、窿缘桉的胚性愈伤组织为亲本材料,采用PEG(聚乙二醇)融合法进行了原生质体融合研究,结果表明:影响桉树原生质体融合的主要因素为:融合液中PEG 克分子量及浓度、Ca2+ 浓度。PEG 以克分子量4 000、6 000 较为合适,所用浓度分别为50% 与35% 时效果最好;融合液中0.4 m ol/L浓度的Ca2+ 的加入有助于原生质聚集体及融合体百分率的提高。融合所用保温温度30℃为宜。融合液中0.4 m ol/L甘露醇的补充有利于原生质与融合体稳定性的保持。异核体核融合率以融合后培养3~5 d 时百分率最高
The embryogenic callus of Eucalyptus urophylla and Eucalyptus urophylla was used as the parent material and protoplast fusion was carried out by PEG (polyethylene glycol) fusion method.
