目的优化接骨木果实中花青素的提取工艺。方法以花青素得率为响应值,分别考察提取溶剂、pH值、提取温度、料液比、提取时间、溶剂浓度等几个因素对接骨木果实中花青素得率的影响。在单因素试验的基础上,采用响应曲面法中的Box-Behnken设计,优化接骨木果实中花青素的提取工艺参数。结果接骨木果实中花青素提取的最优工艺条件为:以69%乙醇(pH=2)为提取溶剂,料液比为1∶40,在49℃下提取30min,花青素得率为2.535mg/g。结论接骨木果实中花青素实测值与响应面模型高度拟合。该方法能合理优化接骨木果实中花青素的提取工艺。
Objective To optimize the extraction of anthocyanin from elderberry fruit. Methods Based on the yield of anthocyanins, the effects of extraction solvent, pH value, extraction temperature, solid-liquid ratio, extraction time and solvent concentration on the yield of anthocyanin in elderberry fruit were investigated. Based on the single factor experiment, the box-Behnken design in response surface methodology was used to optimize the extraction process parameters of anthocyanin in elderberry fruit. Results The optimal extraction conditions of anthocyanin in elderberry fruit were as follows: the extraction solvent was 69% ethanol (pH = 2), the ratio of solid to liquid was 1:40 and the extraction time was 30 min at 49 ℃. The yield of anthocyanins was 2.535 mg / g. Conclusion The results of anthocyanin in elderberry fruit are highly fitted to the response surface model. The method can optimize the extraction process of anthocyanin in elderberry fruit.
