目的:优选大孔吸附树脂纯化头风宁滴丸中菊花的工艺条件。方法:采用高效液相色谱法,以绿原酸比吸附量、洗脱率为指标,对大孔吸附树脂型号、上样量、上样浓度、洗脱剂浓度及用量进行考察。结果:最佳工艺条件为预处理后药液浓度为0.075g·mL-1(以菊花药材计),通过HPD600型大孔吸附树脂柱,上样量为1.2g(生药)/mL树脂,以水洗至流出液近无色,继以4倍树脂体积70%乙醇洗脱。结论:该优选工艺可以较好地纯化富集绿原酸,达到“去粗存精”的纯化效果。
Objective: To optimize the macroporous resin purification Diantan Ning dripping pills chrysanthemum conditions. Methods: The samples of macroporous adsorption resin, loading amount, loading concentration, eluent concentration and dosage were investigated by high performance liquid chromatography (HPLC) with specific adsorption of chlorogenic acid and elution rate as indexes. Results: The optimum conditions for the pretreatment liquid concentration of 0.075g · mL-1 (to daisy herbs), through HPD600 macroporous resin column, the sample was 1.2g (crude drug) / mL resin to Wash until the effluent is nearly colorless, then elute with 4X resin volume 70% ethanol. Conclusion: This process can be better purified chlorogenic acid enrichment, to achieve “to the rough storage of refined ” purification effect.
